Immortalized cell lines derived from mice lacking both type I and type II IFN receptors unify some functions of immature and mature dendritic cells

Abstract: Summary Cells with dendritic morphology obtained from several organs of mice lacking both type I and II IFN receptors were immortalized by a retrovirus and analysed for their phenotype and for their function to induce cognate immune responses in vitro and in vivo. Two cell lines called AG101 (skin) and AG116 (brain) were cloned and analysed in more detail. They constitutively expressed the cell surface markers CD45, CD11b, MHC class II, F4/80, N418, B7-2 and ICAM1 but were CD8-and B220-negative. Cells from bot… Show more

“…Cell analysis was performed using a FACSCalibur ® (Becton Dickinson) and the data were analyzed as previously described ( 23 , 24 ). The following monoclonal antibodies were used: FITC-conjugated hamster IgG group 1, IgG isotype standard anti-trinitrophenol (Cat.…”

“…Using a murine dendritic cell line, JAWS II 18 , and bone marrow-derived DC (BMDC), we tested the effects of short-term (72 h) and long-term (up to 14 d) exposure to SMG upon dendritic cell maturation and capacity to activate peptide-specific CD4 + OT-II T cell hybridomas (TCH) 19 and CD8 + OT-I T cells. Cell surface proteins, such as MHC II, CD80 and CD86, served as markers of maturation.…”

“…It is simple and feasible. The CD11c monoclonal antibody N418 is specific for the integrin αx subunit of αxb2 which was the leukocytic integrin expressed on mouse splenic DC [33,34] . The principle of MACS CD11c+ cell sorting is that the cells are labelled by MicroBeads coupled with CD11c antibodies and passed through a sorting column which is placed in the magnetic field of a MACS sorter.…”
