The effects of an adaptogen-based dietary supplement on stress indicators and stress perception were evaluated in health volunteers using a double-bind, placebo-controlled protocol. Seventy-seven healthy 24-60 year old male and female participants were enrolled, with 74 completing the study. Participants were given a 28-day supply of the supplement or the corresponding placebo and instructed to take 1 oz each morning 30 min. before breakfast. The supplement was a proprietary blend of 10 adaptogens including Eleurtherococcus, Schizandra and Rhodiola extracts, plus B vitamins. On the morning of enrollment, the participants completed a stress perception survey, had blood samples drawn for a highly sensitive C-reactive protein (HS-CRP), cortisol and homocysteine, and returned to the clinic after 28 days to repeat the stress perception survey and blood tests. A 40% reduction (p <0.05) in the number of participants with abnormally elevated HS-CRP in the supplement group compared to the placebo group was observed. Furthermore, there was a 30% reduction in levels of HS-CRP in the supplemented group relative to the placebo group. Only one of the 5 subjects (20%) with abnormal (elevated) serum homocysteine levels remained elevated after 4 weeks of supplementation. In contrast, 4 of the 6 (66%) subjects in the control group remained elevated after 4 weeks. Five stress perception parameters were significantly reduced (10-20%) in the supplemented group compared to two parameters in the placebo group. The results of this study indicate that the adaptogen/vitamin blend is effective at reducing subjective (perception survey) and objective (HS-CRP) stress measures compared to the placebo.

Background/Aims: Autophagy modulation has been considered a potential therapeutic strategy for human chondrosarcoma, and a previous study indicated that salidroside exhibits significant anti-carcinogenic activity. However, the ability of salidroside to induce autophagy and its role in human chondrosarcoma cell death remains unclear. Methods: We exposed SW1353 cells to different concentrations of salidroside (0.5, 1 and 2 mM) for 24 h. RT-PCR, Western-blotting, Immunocytofluorescence, and Luciferase Reporter Assays were used to evaluate whether salidroside activated the TFEB-dependent autophagy. Results: We show that salidroside induced significant apoptosis in the human chondrosarcoma cell line SW1353. In addition, we demonstrate that salidroside-induced an autophagic response in SW1353 cells, as evidenced by the upregulation of LC3-II and downregulation of P62. Moreover, pharmacological or genetic blocking of autophagy enhanced salidroside -induced apoptosis, indicating the cytoprotective role of autophagy in salidroside-treated SW1353 cells. Salidroside also induced TFEB ^(Ser142) dephosphorylation, subsequently to activated TFEB nuclear translocation and increase of TFEB reporter activity, which contributed to lysosomal biogenesis and the expression of autophagy-related genes. Importantly, we found that salidroside triggered the generation of ROS in SW1353 cells. Furthermore, NAC, a ROS scavenger, abrogated the effects of salidroside on TFEB-dependent autophagy. Conclusions: These data demonstrate that salidroside increased TFEB-dependent autophagy by activating ROS signaling pathways in human chondrosarcoma cells. These data also suggest that blocking ROS-TFEB-dependent autophagy to enhance the activity of salidroside warrants further attention in treatment of human chondrosarcoma cells.

Rhodiola kirilowii, a member of Crassulaceae family, grows wildly in Asiatic mountains and is also cultivated in some European countries. Its underground parts traditionally are used for enhance physical and mental performance of the body.In our previous papers we reported immuno- and angio-modulatory effects of aqueous and hydro-alcoholic extracts of radix and rhizome of this plant in mice. In the present work we evaluated the effect of Rhodiola kirilowii water- (RKW) or hydro-alcoholic (RKW-A) extracts and epigallocatechin (one of the polyphenols present in these extracts) given to mice, during pregnancy and nursing period, on the number and localization of CD4+ and CD8+ cells in spleens of adult progeny mice. Previously, we observed several abnormalities in functionality of spleen cells collected from these mice. No differences in CD4+ T cells localisation or numbers were found between all tested mice groups. In contrast, CD8+ T cells localisation and staining were altered in progeny of water or alcohol extract-fed mice. CD8+ T cells were found not only in the PALS but also in the B cell follicle and in the red pulp. Furthermore, CD8+ T cells from T cell zones in the progeny of extract-fed mice showed much intensive staining for CD8 antigen and significantly higher numbers per area in comparison to control mice.
