Immunocytochemical detection of ricin. II. Further studies using the immunoperoxidase method

Abstract: Radio-iodinated ricin was injected into rat muscle in vivo to establish the distribution of the toxin at various time intervals after injection. Injection site muscle and para-aortic lymph nodes were selected for localization of ricin by the immunoperoxidase technique. Sections of snap-frozen tissues were fixed using a variety of methods to establish the best protocol for the immunodetection method. This was found to be with an ether-ethanol mixture. Ricin was detected in tissue at the site of injection taken … Show more

“…In animal studies, ricin has been detected in tissue sections, some tissue specimens, nasal swabs, and fluids by immunologically based methods. 53,92,93 Immunologically based methods have been applied to human and animal fluid specimens in the past and have the potential to measure concentrations as low as 0.1 ng/mL (1.54 pmol/L). [93][94][95][96][97][98] cases, clinicians should collect urine and serum samples and contact their state public health department or CDC for further guidance.…”

“…Subsequently, ELISA was reported for plasma and tissue extract 5 . Griffiths 6 , et al studied immunocytochemical detection of ricin using an immunoperoxidase method on tissue after a ricin injection. Ricin (2 µg) could be detected at the injection site, but was not found by this method in lymphoid tissue, even though radio-labelled ricin was detected in these tissues.…”

“…Destruction of draining LNs following intramuscular injury of the thigh was autologous to the local necrotic lymphadenopathy determined in Markov’s case [11]. In line with these findings, studies on intramuscularly-administered radio-iodinated ricin in rats demonstrated accumulation of radioactivity in muscle draining LNs [31].…”
