Immunocytochemical detection of XIAP in body cavity effusions and washes

Abstract: Body cavity effusions may be the first manifestation of malignancy or of recurrence or relapse. We surveyed effusions and washes for expression of X-linked inhibitor of apoptosis (XIAP), a potent constituent of the inhibitor of apoptosis (IAP) family of proteins. IAPs prevent apoptosis by blocking the activation of caspases, thereby preventing caspase-mediated cell degradation. Elevated expression of XIAP could be an underpinning of relapse and/or resistance to apoptosis-inducing cancer therapy. We performed a… Show more

“…When comparing tumor tissue and peritumoral nontumorous mucosa, greater values for AI, cellular proliferation and p53 immunoexpression in the tumor tissues validated the present results. Greater XIAP immunoexpression in tumor tissues reinforces the findings of other groups (45,46). Increased tumor immunoexpression for SMAC and cIAP-1 fit the models and experimental findings proposed for dynamic relations between the regulatory proteins of caspase-3 activation (27,34,35).…”

“…In addition to Akt, inhibitor of apoptosis protein (IAP) families of proteins also plays a critical role. X-linked inhibitor of apoptosis protein (XIAP) belongs to a group of inhibitors of apoptosis that block the activation of specific caspases and prevent caspasemediated cell degradation (Wu et al, 2005). Specifically, XIAP inhibits caspase 3, 7 and 9 and therefore blocks both intrinsic and extrinsic apoptotic pathways (Kosarac et al, 2011).…”

“…[18][19][20][21] However, immunohistochemical studies specifically addressing XIAP expression in human lung adenocarcinoma and alveolar hyperplasias are lacking. An immunocytochemical study of body cavity effusions and washes performed in our laboratory detected XIAP in eight out of nine metastatic non-small cell carcinomas of pulmonary origin, 22 raising the possibility that XIAP may play a role in the pathogenesis or progression of lung adenocarcinoma.…”
