Immunogenetic Aspects of Nasopharyngeal Carcinoma. IV. Increased Risk in Chinese of Nasopharyngeal Carcinoma Associated With a Chinese-Related HLA Profile (A2, Singapore 2)2

Abstract: The results of this study of 110 Singapore Chinese with nasopharyngeal carcinoma (NPC) and 91 controls confirmed the association between the occurrence of HLA antigen Singapore 2 (Sin2) and NPC in the Chinese population, and indicated that their increased risk for NPC was confined to the joint occurrence of Sin 2 and A2 antigens. These findings suggested that the genotype of importance in susceptibility to NPC is the A2-Sin 2 haplotype.

“…HLA associations have been reported in NPC. With the highest incidence populations, similar HLA associations have been reported among the Chinese from Singapore (Simons et al, 1976;Chan et al, 1983) Malaysia, Hong Kong (Simons et al, 1977), China (Cui & Lin, 1982) and California (Jing et al, 1977). In Singapore, the associations among Chinese NPC were with A2 BW46 among older onset NPC and AW19 B17 among total NPC but this association was especially strong among younger onset NPC (Chan et al, 1983).…”

“…In southern Chinese and other Asian populations, HLA-A2-B46 (252,(277)(278)(279)(280)(281)(282)(283)(284) and B17 (281,282,(285)(286)(287) were generally associated with a 2-to 3-fold increase in NPC risk. In contrast, 30% to 50% lower risk of NPC was found in association with HLA-A11 in both Chinese and Whites (277, 281-283, 287, 288), B13 in Chinese (279,282), and A2 in Whites (288,289).…”

“…The epidemiological pattern may be because of genetic susceptibility correlated with certain Chinese-related HLA antigen profiles and/or to environmental factors (the consumption of salted fish or exposure to fumes, smoke, and chemicals from the occupational environment; Refs. 197,[204][205][206][207][208][209][210][211].…”
