“…The genome inactivation was done upon a controlled UV treatment of the cultured virus sample. We chose a suitable UV exposure or dose that affected the viral genome without altering the structural integrity of the virus as reported by recent literature on the UV disinfection of SARS-CoV-2 . “…10 mL of viral supernatant was UV-inactivated using a UV chamber in a UV crosslinker at wavelength 254 nm for ∼4 min (Amersham Biosciences) set on the energy mode at 2700 × 100 μJ energy. For UV disinfection, we kept the power in the UV chamber within <500 μW/cm 2 , which can only damage the viral genome without any aberrant effect on the morphology of virions as also previously described by many researchers. ,, The UV-treated viral samples were concentrated using a 100 kDa concentrator to remove any free proteins.
