Immunohistochemical characterization of neurons and neuronal processes in the dorsal vagal nucleus of the pig

Abstract: The vagus nerve is responsible for efferent and afferent innervation of the gastrointestinal tract. The efferent fibres originate from neurons located in the dorsal motor nucleus of the vagus (DMX) and control mainly the gastric motor and secretory function. The main goal of our study was to examine expression of ChAT, SP, LENK, NOS and CART in the neuronal matrix of the porcine DMX. Double-labeling immunofluorescence revealed plenty of ChAT-IR neuronal cell bodies and fibres distributed throughout the nuclear … Show more

“…Identification of the SP NK 1 receptor on vagal efferent somata provides direct confirmation of this pathway (Plata-Salaman et al 1989; Le Brun et al 2008). Possible physiological implications of the SP, LENK, CART, GAL, NOS, and VIP immunopositive processes in the regulation of stomach function have been outlined in our former reports (Gańko and Całka 2013; Gańko et al 2013). …”

“…ChAT, as a primary motor fascicles marker, is widely reported to be highly expressed in human facial motor neurons and rat facial motor neurons [ 38 , 39 ], with high concentrations in motor neurons in both the central nervous system (CNS) and peripheral nerve axons [ 40 , 41 ]. A key feature of motor neuron development and function is the expression of the acetylcholine biosynthetic enzyme, ChAT [ 42 , 43 ]. Yuan et al used ChAT as an enzymatic marker in immunohistochemistry staining to detect motor axons and used NF200 to detect sensory axons and showed that the sciatic nerve contains both motor and sensory axons [ 44 ].…”

“…Neurons localized in the DMV, project to the upper gastrointestinal tract including the liver, whereas those located in the NA project to the esophagus and the cardiorespiratory system. Despite integration of peripheral and central signals concerning gastrointestinal processes, the preganglionic parasympathetic cells of the DMV also directly influence gastrointestinal activities (12).…”
