“…The predominant effect of AVP is one of syn ergism with CRF on the release of adrenocorticotropic hor mone (ACTH) from pituitary corticotropes [8,17]; it has been co-localized with CRF in neurosecretory vesicles in the median eminence [22] and, in the adrenalectomized rat, in the perikarya of the parvicellular cells in the paraventri-cular nucleus of the hypothalamus [11,13,23], Indeed, CRF itself was thought to be a neurophysin-associated com pound before its isolation in 1981 [21], We have therefore studied the ontogeny of AVP in addition to that of CRF, to determine whether the two are expressed in parallel over the course of development.…”

“…This illus¬ trates that the AVP involved in ACTH release may arise directly from the hypothalamus in true neurosecretory fashion, rather than originating indirectly from the posterior lobe (Porter, Eskay, Oliver et al 1978), thus substantiating its role as a true CRF. This idea is amply supported by the extent of VP and neurophysin immunostaining in the zona externa of adrenalectomized and dexamethasone-treated animals (Watkins, Schwabadel & Bock, 1974;Zimmerman, Stillman, Recht et al 1977 (Linton et al 1983). It is not surprising that the CRF RIA was unable to detect CRF-41-like immunoreactivity in the effluent as the sensitivity of the RIA developed against synthetic oCRF is 100 ng/1 and it detects a total of only 700 pg per hypothalamus.…”

“…Lesions of the paraventricular nuclei abolished this median eminence vasopressin, suggesting that vasopressin fibers in the median eminence originate from the paraventricular nuclei (215). Vasopressin and neurophysin in the median eminence increased markedly after adrenalectomy; this increase was reversed by treatment with glucocorticoids (215,217,218).…”
