Immunohistochemical Detection of Dendritic Cell Markers in Cattle

Abstract: Dendritic cells (DCs) are ''professional'' antigen-presenting cells with a critical role in the regulation of innate and adaptive immune responses and thus have been considered of great interest in the study of a variety of infectious diseases. The objective of this investigation was to characterize the in vivo distribution of DCs in bovine tissues by using potential DC markers to establish a basis for the study of DCs in diseased tissues. Markers evaluated included MHCII, CD208, CD1b, CD205, CNA.42, and S100 … Show more

“…It was detected that IHC studies were carried out in the lymph node of rat with S100 protein [6], Romero-Palomo et al [13] has employed polyclonal rabbit anti-S100 primary antibody as dendritic cell marker in the lymph node of cattle. There is also a comparative study on S100 protein-immunoreactive cells in the lymph nodes of guinea pig, dog, cat, horse, pig, goat and cow [14].…”

“…Thymic DCs are considered the major cell population responsible for the process of immunologic central tolerance, by participating in the deletion of self‐reactive developing T cells (Ardavin, ) and the induction of natural regulatory T cells (Proietto et al., ). For this reason, we studied this type of cells by immunolabelling against two molecules (CD1b and CD208) previously reported as DC markers in cattle (Romero‐Palomo et al., ).…”

“…Antibodies against CD205 and CD208 antigen were applied for the detection of DCs, as previously reported in cattle (Romero-Palomo et al, 2013), sheep (Åkesson et al, 2008) andhumans (de Saint-Vis et al, 1998;Geijtenbeek et al, 2000). Only CD208 (synonym dendritic cell lysosomal-associated membrane protein [DC-LAMP]) revealed specific immunoreaction on few histiocytic cells in camelid tissue ( Fig.…”
