Immunohistochemical Localization of Collagen Type XI α1 and α2 Chains in Human Colon Tissue

Abstract: In previous studies, collagen XI mRNA has been detected in colon cancer, but its location in human colon tissue has not been determined. The heterotrimeric collagen XI consists of three alpha chains. While it is known that collagen XI plays a regulatory role in collagen fibril formation, its function in the colon is unknown. The characterization of normal human colon tissue will allow a better understanding of the variance of collagen XI in abnormal tissues. Grossly normal and malignant human colon tissue was … Show more

“…Additionally, COL11A1 epithelial expression has also been demonstrated at the protein level, not only in breast tissue [28] but also in colorectal epithelium [25]. The co-expression and concomitant up-regulation of COL11A1 with COL5A2 has previously been shown in colorectal cancer compared to its precursor lesion [26,27], which is in agreement with our data that showed up-regulation of both these genes in IDC relative to DCIS.…”

“…[23,24]. COL11A1 encodes a minor collagen found in many tissues; its expression has been shown to be dysregulated in cancers such as breast and colon [25][26][27][28]. COL17A1 is a transmembrane protein expressed in normal breast and involved in cell adhesion and SOX10, a transcription factor of the SRY family of the high mobility group box family, is known to be expressed in normal myoepithelial cells [29].…”

“…Previous observations on the expression of COL11A1 in other cancers [40-44] were limited to differential gene expression analyses validated by quantitative RT-PCR, Northern blots and/or in situ mRNA hybridization. Recently, the expression of collagen XI in both normal and malignant human colon tissue has been assessed by immunohistochemistry [45]. …”

“…Sections were deparaffinized in xylene, rehydrated through graded ethanol, and incubated for 30 minutes in 0.01 mg/mL hyaluronidase (Sigma-Aldrich Corporation, St. Louis, MO) [11]. Primary antibody to collagen α1(XI) was diluted 1:400 and applied to tissue section for 1 hour at 25°C [12]. An antirabbit secondary antibody conjugated to horseradish peroxidase was applied and incubated for 30 minutes, followed by development with substrate chromogen (DakoCytomation, Dako, North America).…”
