Immunohistochemical Localization of SERCA2 in the Ameloblasts of Rat Incisors

Abstract: Sarcoplasmic or endoplasmic reticulum Ca 2+ ATPase (SERCA2) was localized in the ameloblasts of rat incisors by immunohistochemistry. Secretion ameloblasts and maturation ameloblasts, including both ruffle-ended and smooth-ended cells, were labeled with anti-SERCA2 antibodies, with maturation ameloblasts showing stronger binding than secretion ameloblasts. The pattern of labeling was cytoplasmic and diffuse. Papillary layer cells were not labeled with anti-SERCA2 antibodies. These results suggest that the SERC… Show more

“…For goat antibodies, donkey anti-goat IgG conjugated with biotin (Santa Cruz) diluted 1:100 was used in place of the link antibody of the LSAB2 Kit. Fluorescent images were visualized using an Olympus AX80 fluorescence microscope equipped with a CCD camera (Quantix KAF1401E, Photometrics, Tucson, AZ) and using MetaMorph software (Universal Imaging, Downingtown, PA) [12]. In quadruple labeling, four cubes for epifluorescence microscopy were used: U-MNUA (Olympus, Tokyo, Japan) filtered through 360-370 nm for excitation and 420-460 nm for emission, U-MNIBA (Olym-…”
