Immunohistochemical study of mucin expression in periampullary adenomyoma

Abstract: A combined evaluation of IHC findings may be effective in the detection of AM and AMH, and also in distinguishing benign periampullary lesions, such as AM and AMH, from ampulla of Vater adenocarcinoma, thus avoiding excessive surgery.

“…For MUC3 staining, the sections were treated at 100°C for 10 min in 0.01 M citrate buffer at pH 6.0 and then reduced with 0.01 M dithiothreitol in 0.1 M Tris/HCl buffer (pH 8.0) for 30 min at room temperature and alkylated with 0.025 M iodoacetamide in 0.1 M Tris/HCl buffer (pH 8.0) for 30 min [10], [15]. They were then incubated with mMUC3-1 at 4°C for 16 h and stained by the avidin-biotin complex method.…”

“…For example, normal pancreatic tissue mainly expresses MUC1 and MUC6, while ampullary adenocarcinoma mainly expresses MUC1 and MUC5AC [10], along with MUC6 and MUC2 [3,11,12]. Combined detection of mucins has been used to diagnose and differentiate pancreatic ductal adenocarcinoma and ampullary adenocarcinoma [11-15]. …”

“…Immunohistochemical stainings were done by an immunoperoxidase method using the ABC complex as described previously [2,6,9,15] . For the retrieval of MUC4, the sections were treated in a water bath at 80 ° C for 20 min in 0.01 M citrate buffer (pH 6.0).…”
