Immunologic properties of differentiated and undifferentiated mesenchymal stem cells derived from umbilical cord blood

Abstract: The expression of immunogenic markers after differentiation of umbilical cord blood (UCB)-derived mesenchymal stem cells (MSC) has been poorly investigated and requires extensive in vitro and in vivo testing for clinical application. The expression of human leukocyte antigen (HLA) classes on UCB-derived MSC was tested by Fluorescence-activated cell sorting analysis and immunocytochemical staining. The undifferentiated MSC were moderately positive for HLA-ABC, but almost completely negative for HLA-DR. The MSC … Show more

“…Furthermore, host-derived pro-inflammatory cytokines can impair the capability of MSCs to differentiate into bone, fat, and cartilage lineages [see ( 94 )]. However, even after MSCs have differentiated into chondrocytes, they can exert immunosuppressive effects ( 108 , 109 ).…”

“…Thus, allogeneic stem cells derived from healthy humans may be an alternative for treating ONFH. Interestingly, there is evidence for the accumulation of low-immunogenicity MSCs, which allows the MSCs to be transplanted between human leukocyte antigen (HLA)-incompatible individuals [ 34 ]. hUCMSCs may be a good candidate for this approach, because umbilical cord (UC) collection is easy and ethically feasible.…”

“…With regard to the effect of MSC differentiation on the cell surface profile of the cells, there are conflicting results presented in different studies. For example, Li et al describe an upregulation of HLA-DR on MSC after hepatocyte differentiation ( 30 ), while several other studies report no change in cell surface proteins after chondrogenic ( 31 ), myogenic ( 32 ), and insulin-producing cell ( 33 ) differentiation.…”
