Immunomagnetic Capture of Faecalibacterium prausnitzii Selectively Modifies the Fecal Microbiota and Its Immunomodulatory Profile

Abstract: There is increasing interest in deciphering the contribution of gut microbiota species to health and disease amelioration. The approach proposed herein provides a novel and affordable strategy to probe deeply into microbiota-host interactions by strategically modifying the relative abundance of specific gut microbes, hence facilitating the study of their contribution to a given trait of the microbiota.

“…Notably, a recent study provided a method that can be a potential reference. Marcos et al [81] prepared specific polyclonal antibodies against the surface of Faecalibacterium prausnitzii M21 and captured Faecalibacterium prausnitzii from the human intestinal microbiota by combining immunomagnetic separation with flow cytometry. However, it is important to note that the residue of immunomagnetic beads on the bacterial surface may lead to bacterial adhesion and, thus, affect the accuracy of flow sorting.…”

“…Inflammatory bowel disease (IBD) is a chronic inflammation-related condition affecting the gastrointestinal tract, influenced by a combination of factors including diet, environment, genetics, and immune dysregulation. Over the past decade, the improvement of living standards has led to an increased consumption of high-fat and low-fiber diets, leading to a higher incidence of diseases such as colitis, colorectal cancer, and metabolic disorders. , However, traditional pharmacological and biological therapies have side effects and may lead to drug dependence. , Therefore, it is essential to explore new natural approaches that are devoid of side effects for the treatment of colitis . Dextran sulfate sodium salt (DSS) is often used in models of colitis, and current studies have mainly linked DSS to increased intestinal permeability, disruption of the intestinal mucosal barrier, upregulation of cytokines, activation of pathways, and dysbiosis of the gut microbe.…”
