Immunomodulatory Properties of Adipose-Derived Stem Cells Treated with 5-Azacytydine and Resveratrol on Peripheral Blood Mononuclear Cells and Macrophages in Metabolic Syndrome Animals

Abstract: Endocrine disorders, including equine metabolic syndrome (EMS), are a serious issue in veterinary medicine and horse breeding. Furthermore, EMS was shown to affect the cytophysiological properties of adipose-derived stem cells, reducing their therapeutic potential. However, it was shown that those cells can be rejuvenated while using a combination of two chemicals: 5-azacytydine (AZA) and resveratrol (RES). In the present study, we decided to evaluate the immunomodulatory properties of AZA/RES-treated adipose-… Show more

“…Indeed, MSC-derived vesicles were shown to have positive impact on tissue repair via various pathways, mainly those associated with cell death and senescence, for instance resulting from ischemic injury [47]. It is also emphasized that therapeutic effect of mesenchymal stem cells relies on their paracrine effect and immunomodulatory properties [47,48], due to this fact various cell-free therapies based on MVs implantation are being evaluated [49].…”

“…Importantly, these ASCs can be quickly isolated using differential centrifugation in the operating room, making ASCs a viable candidate for use in any point-of-care procedure. In addition, like MSCs, ASCs secrete a multitude of therapeutic growth factors and cytokines that are anti-inflammatory and immunomodulatory [27][28][29][30][31][32][33].…”

“…Analysis of gene expression was performed after the fifth day of differentiation as described elsewhere [39]. Total RNA was extracted using the phenol-chloroform method as originally described by Chomczynski and Sacchi [40].…”
