Impact of liver-specific GLUT8 silencing on fructose-induced inflammation and omega oxidation

Abstract: Excessive consumption of high-fructose diets is associated with insulin resistance, obesity, and non-alcoholic fatty liver disease (NAFLD). However, fructose differentially affects hepatic regulation of lipogenesis in males and females. Hence, additional studies are necessary in order to find strategies taking gender disparities in fructose-induced liver damage into consideration. Although the eighth member of facilitated glucose transporters (GLUT8) has been linked to fructose-induced macrosteatosis in female… Show more

“…GLUT8 has recently been reported to be involved in fructose-mediated NAFLD in the female mouse liver [ 44 ]. TM4SF5 promotes NASH-associated fibrosis [ 17 ] and is involved in the development of CCl 4 -mediated fibrosis in the mouse liver [ 16 ].…”

“…Yet, whether CD53 mechanistically participates in the adaptive metabolic effects, which we previously demonstrated in GLUT8-deficient mice ( e.g. , protection from NAFLD and insulin resistance ( 32 , 33 , 38 , 44 , 53 , 70 , 71 , 72 )), remains to be fully determined. Nevertheless, we introduced a novel scaffolded trehalose polymer, which is a unique composition of matter, and water-soluble compound that induces hepatocyte fasting–like signaling responses, and inhibits CD53 expression in unperturbed, fructose-, and FA-treated hepatocytes to an equal or greater extent than trehalose.…”

“…The biological significance of this route occurring in the liver and other organs is the generation of more soluble acids that are cleared by the kidneys, especially for those on low-carbohydrate or ketogenic diets. Despite the notion that ω-oxidation is catalyzed by enzymes that induce ROS and NAFLD progression [ 55 ], other studies consider it a rescue pathway for fatty acid oxidation disorders [ 56 ].…”
