Impact of selected novel plant bioactives on improvement of impaired gut barrier function using human primary cell intestinal epithelium

Abstract: Gut barrier function is compromised in the obese state. The N-trans caffeoyltyramine (NCT) and N-trans feruloyltyramine (NFT), two naturally occurring bioactive compounds in hemp hulls,  identified using in silico approaches, have the potential to improve gut barrier function and their effects were studied here in vitro. Proliferative human transverse colon epithelial cells were plated and co-cultured with tumor necrosis factor (TNF) along with NCT, NFT or NCT/NFT (2.2 ratio) post-differentiation, over a 48-ho… Show more

“…5 Hemp hulls are also rich in bioactive phenolics, which enhance gut barrier function in vitro. 6 Bio gut fibre (BGF) is a minimally processed fibre ingredient from hemp hulls and is generally recognized as safe (GRAS status, U.S. FDA 2021a). We hypothesized that BGF reduces intestinal permeability in BAD.…”

“…In addition to IDF, hemp hulls contain lignanamides, including cannabisin F-type (34-50 mg/100 g DW) and grossamide-type (76-292 mg/100 g DW) [32]. These bioactive compounds are formed using hydroxycinnamic acid amides as intermediates and have in vitro antioxidant and anti-inflammatory activity [60,61]. Blood orange fruits are rich in bioactives; seeds were the highest in limonoids, but peels had the highest flavonoid content.…”

“…Fibers such as galacto-oligosaccharides (GOS), fructo-oligosaccharides (FOS), and resistant starch (RS) reach the colon and are fermented by bacteria into short-chain fatty acids (SCFAs) 5 that can strengthen gut barrier function and mitigate enteric inflammation 6,7 . Consequently, there is growing interest in understanding the interactions between complex dietary fiber and the gut microbiome, but most studies to date have focused on dietary fiber available through whole foods 8,9 or structurally repetitive fibers such as GOS and FOS 10,11,12 .…”
