In ectothermic animals, heat seems to be a determining factor because it influences many vital activities such as locomotion, the ability to escape, feeding, and reproduction, among others. In aquatic environments, physical characteristics of water prevent small ectotherms from thermoregulating and therefore it is expected that their body temperature remains similar to water temperature. Throughout its distribution and annual cycle, the Pyrenean newt (Calotriton asper) is exposed to a wide range of water temperatures that affect its biological and ecological traits like the standard metabolic rate, oxygen consumption, activity period and growth pattern. This study analyses the relationship between the cloacal and water temperatures in a Pre-Pyrenean population of C. asper in two periods with well differentiated water temperatures (July and September). The aims are to establish if there are differences between sexes in cloacal temperature, whether reproductive activity modifies cloacal temperature, and to analyse the degree of thermoconformity of the species. The results indicate that cloacal temperature depends on the water temperature, corroborating the idea that C. asper is mainly a thermoconformer, as it corresponds to an aquatic ectotherm. However, the cloacal temperature of newts was slightly higher than water temperature. In individuals in amplexus, cloacal temperature was significantly higher than in specimens that are not in this mating position, and in July, females showed much greater dispersion in cloacal temperature than males, which is probably related to spawning behaviour.

Gathering ecological knowledge about species with cryptic behaviour can be challenging and strongly depends on human, financial and logistical resources. Yet, this is essential for the conservation of threatened and/or elusive species and wildlife management. The use of DNA tools is emerging as an efficient solution to overcome these constraints. Here, we developed specific primers and a droplet digital PCR (ddPCR) protocol for the quantitative detection of the endemic amphibian Calotriton asper in freshwater and fish faeces. After successful in silico validation of the developed primers, we tested their validity in the wild. First, using an environmental DNA approach, we confirmed the presence of C. asper in eight out of 13 sites deemed climatically suitable for the species, although it had never been observed there before. Then, we sampled brown trout (Salmo trutta) living in sympatry with C. asper and, through fish faeces analyses, confirmed that brown trout are predators of C. asper. These molecular tools proved efficient for monitoring (i) elusive species such as C. asper in the wild, allowing for better characterization of their current distribution and habitat use, and (ii) predation events by fish, which might be a significant pressure on low‐density and isolated C. asper populations. We provide a valuable, low‐cost and complementary tool for environmental managers and scientists that should help improve the development of fine‐tuned conservation and management plans for C. asper.

Whether and how poikilothermic animals change their thermal performance to cope with global warming are crucial questions to predict the future of biodiversity. Intraspecific comparison among populations that occur in different climatic zones can provide insight into how poikilotherms may alter their thermal performance under a particular climatic event. We compared populations of the Asiatic toad (Bufo gargarizans) from two altitudinal zones (3239 and 926 m above sea level) to explore variations of post-exercise hypothermia, which can lead to lower temperature preference than normal conditions. Common garden experiment was also employed to test plasticity of hypothermic performance in adult toads. As results, exhaustive exercise induced measurable reduction in body temperature for both populations. Furthermore, high-altitude population experienced larger reduction in body temperature than low-altitude conspecifics in both original habitat and common garden conditions. Therefore, low-altitude toads may to enhance their hypothermic reaction if they shift their ranges to higher altitudes to survive warming climate; However, the relatively limited plasticity of hypothermic performance may constraint their adaptative process.
