Improved technique of heterotopic auxiliary rat liver transplantation with portal vein arterialization

Abstract: We developed a standardized and improved technique, which can be used for experiments of regeneration and inter-liver competition in auxiliary liver transplantation. Furthermore, this technique is suitable for the investigation of the influence of portal vein arterialization and portal hyperperfusion on liver microcirculation, function, and morphology.

“…Subsequent to this, various ALT models in rats have been developed ( 15 17 ). Of these models, the APHLT with PVA model in rats was developed by Schleimer et al ( 7 ); one of advantages of this model is that portal venous blood flow is controlled by the connected stent. This model is simple and easy to perform.…”

“…For these reasons, several clinical studies using APHLT with PVA have been conducted in recent years ( 4 6 ); however, the graft liver has been found to fail in long-term survival. To solve this problem, Schleimer et al developed a rat model of APHLT with PVA ( 7 ). In this model, the graft portal vein is connected by a stent with 0.3 mm inner diameter to the recipient’s right renal artery.…”

“…Heterotopic auxiliary and reduced (30% of the total volume) rat liver transplantation with portal vein arterialization was performed in adult, male Sprague Dawley rats using a described technique . The engraftment was done without performing the hepatic artery anastomosis.…”

“…Heterotopic auxiliary and reduced (30% of the total volume) rat liver transplantation with portal vein arterialization was performed in adult, male Sprague Dawley rats using a described technique. (23) The engraftment was done without performing the hepatic artery anastomosis. Macroscopy appearance, color, and consistency of the engrafted livers after reperfusion was identical to the native livers ( Fig.…”

“…The animal models in groups A and B were prepared based on methods previously described ( 4 , 7 ). The animal model in group C was established on the basis of an improvement of the methods described by Schleimer et al ( 8 ). In brief, the recipient underwent a 68% liver and right kidney resection.…”
