Improvement of Anaerobic Operation and Isolation Culture on Hydrogen-Producing and Fermentative Bacteria

Abstract: Isolation and culture of hydrogen-producing and fermentative bacteria is an important foundation on biohydrogen production process. There are complicated operation and composition in present anaerobic techniques and culture media. Hungate technique was improved and plate of culture bottle was established by comparing anaerobic methods and bacterium growth. Isolation and enrichment culture media were confirmed by the test of different composition and the species and amount of hydrogen-producing and fermentative… Show more

“…After sterilization under conditions of high pressure at 115℃ for 30 min, the isolated fermentation bacteria were cultured at 120r/min, and the culture temperature was 37℃. An improved Hungate rolling tubes technique was utilized for preparing the aseptic water, culture media, isolating culture and bacteria operation [7] .…”

“…Nowadays, scientists only got some species that come from Clostridium genus and Enterobacter genus. Their genetic background is narrow and could not be used in practice [3,17]. For widening genetic basis of the bacterium and discovering the new species, we isolated some high efficient biohydrogen production anaerobic bacterium and made phylogenetic analysis.…”
