In planta production of a candidate vaccine against bovine papillomavirus type 1

Abstract: Bovine papillomavirus type 1 (BPV-1) is an economically important virus that induces tumourigenic pathologies in horses and cows. Given that the BPV-1 L1 major coat protein can self-assemble into highly immunogenic higher-order structures, we transiently expressed it in Nicotiana benthamiana as a prelude to producing a candidate vaccine. It was found that plant codon optimization of L1 gave higher levels of expression than its non-optimized counterpart. Following protein extraction, we obtained high yields (18… Show more

“…The sequences were codon-optimized for the N. benthamiana genome to increase the yields of expressed protein [28]. The synthetic gene was used as template for insertion of a 6x His-tag at either the N- or C-terminus by the PCR DNA amplification method (see Methods).…”

“…The idea of developing a vaccine to combat BPV began with the infection of Shope papillomas extract in the 1940 decade (Shope, 1937). Since then, different vaccine models were proposed (Jarret et al , 1991; Gaukroger et al , 1996; Góes et al , 2008; Love et al , 2012; Mazzuchelli-de-Souza et al , 2013). However, none of them became a commercial product.…”

“…Although the CPMV- HT system has been used to generate VLPs from a number of other viruses, such as Hepatitis B core particles [17] and Human [27] and Bovine [28] Papillomavirus particles, all of which consist of a single type of subunit, the system was recently used to reconstruct fully assembled and immunogenic Bluetongue virus (BTV) VLPs [29 •• ]. This required not only the simultaneous expression of four structural proteins but was optimised by varying the expression levels between the proteins to favour the accumulation of fully assembled particles over assembly intermediates (Fig.…”
