In vitro and in vivo studies on salifluor/PVM/MA copolymer/NaF combination as an antiplaque agent

Abstract: Salifluor (5-n-octanoyl-3'-trifluoromethyl-salicylanilide), a broad spectrum antimicrobial agent, was investigated for its ability to inhibit dental plaque formation. A combination of salifluor with PVM/MA copolymer and NaF was optimized for its antiplaque effect in mouthrinse and dentifrice formulations based on a series of both laboratory and clinical studies. It was found that salifluor, a highly hydrophobic compound, could not be adequately solubilized with the conventional amount of sodium lauryl sulfate … Show more

“…It also enhances triclosan reservoirs within plaque biofi lm. 54,55,72,78 The presence of the PVM-MA copolymer appears to be necessary in a triclosan/fl uoride toothpaste formulation in order for periodontal health benefi ts to achieve statistical signifi cance over a prolonged study period. For example, statistically significant reductions in the Gingival Index 79 and the severity of gingivitis were noted only for subjects randomised to the triclosan/ PVM-MA copolymer fl uoride toothpaste in a six-month double-blind study of unsupervised brushing twice daily (N = 194).…”

“…PVM/MA copolymer (trade name Gantrez) has been utilized as a delivery system for triclosan. PVM/MA copolymer promotes uptake of triclosan by enamel and buccal epithelial cells (Nabi et al, 1989).…”

“…6 The use of copolymers to improve active agent delivery is well established. 7 Methoxyethylene and maleic acid (Gantrez) are copolymers that have been proven to increase triclosan retention in the oral cavity, but the precise mechanism of this enhancement is still unknown. 12,13 Some authors have reported that, when used in combination with Triclosan, Gantrez clearly enhances the uptake of this antibacterial agent by teeth and soft tissues.…”
