In vitro and in vivo evaluation of Ro 09-1428, a new parenteral cephalosporin with high antipseudomonal activity

Abstract: Ro 09-1428, a new parenteral cephalosporin with a catechol moiety attached at position 7 of the cephalosporin ring, showed high in vitro activity against Escherichia coli, Kkbsielia pneumoniae, Proteus mirabilis, Proteus vulgaris, and Streptococcus pyogenes, with MICs for 90% of strains tested (MIC9s) of c0.39 ,ug/ml.MorganeUa morganii, Providencia reftgeri, Citrobacterfreundii, Haemophilus influenzae, Staphylococcus aureus, and Streptococcus pneumoniae were inhibited with MIC90s of <3.13 ,ug/mi. Serratia marc… Show more

“…These were mainly cephalosporins, carbacephalosporins, carbapenems or monobactams with one or two catecholate or mixed catecholate hydroxamate moieties. (Arisawa et al 1991;Ghosh et al 1996;Chang et al 1997;Roosenberg et al 2000). Antibacterial activity of the SDCs was increased when compared to those of the nonmodified drugs under conditions of iron starvation, preferentially against P. aeruginosa strains.…”

“…An increase in the production of alginate has been observed when P. aeruginosa PAO 568 (the muc-2 cells) is cultivated in a medium with an increase in the NaCl concentration (18). Antibiotics continue to play a profound role in the care of patients with CF, and antibiotics with excellent activities against P. aeruginosa are being found (5). It is also interesting that biofilm cells of P. aeruginosa have been shown to produce 32-fold more 1-lactamase than cells of the same strain grown planktonically (22).…”

“…This "Trojan Horse" strategy (4,5) is exemplified by natural siderophore-antibiotic conjugates such as salmycins, albomycins (6), ferrimycins, and microcins (7) produced by various microorganisms. Inspired by these structures, synthetic siderophore conjugates have been designed in the past mainly as a combination between ␤-lactams and a catechol substituent to promote uptake through dedicated siderophore receptors (8)(9)(10)(11).…”
