In Vitro Antibacterial Activity of Amikacin and Ticarcillin, Alone and in Combination, Against Pseudomonas aeruginosa

Abstract: In vitro antimicrobial susceptibility studies using amikacin and ticarcillin, alone and in combination, were performed on 20 strains of Pseudomonas aeruginosa . All strains were susceptible to amikacin, and ticarcillin was active against 16 of the 20 strains. Enhanced anti-pseudomonas activity could be demonstrated with the combination of amikacin and ticarcillin.

“…Although antimicrobial synergy appears to be best established for ␤-lactam-aminoglycoside combinations, similar data on synergistic activity have emerged for combinations of ␤-lactams and fluoroquinolones (14,50,77,96,99,132,168,183,189,205,252,253). In vitro synergy between ␤-lactams and fluoroquinolones against Gram-negative organisms has ranged from 17% to 82% (96,119,168).…”

“…While more than one antimicrobial is frequently needed to cover the entire spectrum of suspected microorganisms in severe sepsis and septic shock, combination antimicrobial therapy is the use of more than one antimicrobial to target the same microorganism through different pharmacologic mechanisms of action to achieve an additive or synergistic killing effect leading to more rapid clearance of the infection [6971]. Historically, evidence supporting antimicrobial synergism to this effect can be found with β-lactam/aminoglycoside combinations [7275]. Similar findings have also been demonstrated for β-lactam/fluoroquinolone combinations [7679].…”

“…Bactericidal synergy was felt to be present if the MBCs determined for both single antibiotics each fell fourfold or greater when these agents were used in combination [20]. To verify the occurrence of bac tericidal synergy, timed-kill assays were also per formed.…”

“…Minimum inhibi tory concentrations (MICs), minimum bactericidal concentrations (MBCs) and the bactericidal synergy of amikacin and ticarcillin for this PA strain were de termined by the checkerboard microtiter technique [20]. Overnight MH broth cultures of PA-96 were ad justed to a McFarland No.…”
