In vitro biosynthesis of juvenile hormone in larval honey bees: Comparison of six media

Abstract: We have formulated a tissue culture medium based on the components of larval honey bee hemolymph. Using an in vitro radiochemical assay to measure juvenile hormone biosynthesis, we compared our larval-based medium to four commercially available media (Grace's, Medium-199; Shields and Sang M3, and Minimum Essential Medium), and a medium based on adult honey bee hemolymph. All media were formulated without methionine. There was no significant difference in the amounts of juvenile hormone produced by the larval m… Show more

“…Ovaries were dissected in honey bee saline, and the ovarioles were individualized in honey bee culture medium (Rachinsky and Hartfelder, 1998) before tracheae and the peritoneal sheath were manually removed as far as possible with the aid of number 5 watchmaker forceps (Dumont). The individualized ovarioles were fixed for 2 h in a glutaraldehyde/paraformaldehyde (2%/2%) mix in cacodylate buffer (50 mM, pH 7.4).…”

After two brief rinses in fresh medium, the ovarioles were individualized and cleared of trachea and peritonial sheath before fixation (overnight at 4…”

“…For probing the ecdysone response in ovaries in vitro, batches of 50 L5F3 worker larvae per sample were dissected and their ovaries were incubated for 6 h at 34.5°C in 250 µL larval honey bee medium (Rachinsky and Hartfelder, 1998) supplemented with makisterone A (final conc. 5 × 10 -7 M) (Hartfelder et al, 1995).…”
