In Vitro Development of Bovine Embryos after Fertilization using Semen from Different Donors

Abstract: Contents:The aim of this study was to determine whether the semen donor andlor heparin concentration injluences the rate of fertilization of bovine follicular oocytes and their subsequent embryonic development in vitro. Frozen-thawed semen from five highly fertile bulls was treated with one of four concentrations of heparin (0.5,l .O, 2.0 and 5.0 kglml) on a 5 x4 factorial basis in an IVM-NF programme. Zygotesloocytes were cultured in frozenthawed bovine oviduct cell-conditioned medium for 6 days. The use of s… Show more

“…These results agree with the work of Shamsuddin and Larsson [21], who have demonstrated that the use of different bulls during IVF leads to different embryo developmental rates until the fourth cellular cycle (16 cells). However, when this particular developmental stage, which corresponds to embryonic genome activation, is bypassed, embryo development is similar among bulls until the morulae/blastocyst stage.…”

“…However, many authors indicated the benefit of using IVP data to estimate in vivo bull fertility [10, 11, 21, 24]. Advances in bovine IVP embryos systems have allowed the relationship between in vivo bull fertility and IVP outcomes to be examined [12, 2527].…”

“…When considering the use of fertility tests, the relationship between age and fertility should also be considered. In most laboratory tests, as well as in most published studies, a single test of a semen sample was used to represent the long‐term or working‐lifetime fertility of a given bull ( Hillery et al ., 1990 ; Marquant‐Le Guienne et al ., 1992 ; Shamsuddin & Larsson, 1993). Our former work ( Zhang et al ., 1997 ) has shown that within‐sire variation in fertility exists between ejaculates.…”

“…This selection implies both a selection of semen donor and individual sperm cells within the ejaculate. The influence of the bull on the Outcome of the IVF and on the further development of the fertilized oocytes have been documented in several studies (Shi et at., 1990;Barandi et at., 1993;Shamsuddin & Larsson, 1993). Efforts have been made to correlate the result of IVF/WP with the results from artificial insemination (AI) to use in vitro techniques as a mean to predict the fertility of bulls after Al.…”

“…Efforts have been made to correlate the result of IVF/WP with the results from artificial insemination (AI) to use in vitro techniques as a mean to predict the fertility of bulls after Al. The results of these studies have, however, been contradictory (Marquant-Le Guienne et at., 1990; Kawaguchi, 1992;Shamsuddin & Larsson. 1993).…”
