In vitro effect of karathane LC (dinocap) on human lymphocytes

Abstract: Karathane LC (active ingredient dinocap), a contact fungicide and a non-systemic acaricide was investigated for its ability to induce chromosome aberrations (CAs) and sister chromatid exchanges (SCEs) in cultured human lymphocytes of peripheral blood. In addition to the cytogenetic analysis, the effect of karathane LC on the cell proliferation kinetics (CPK) by the replication index (RI) was studied. The mitotic index (MI) was also determined to detect the cytotoxic effect. Lymphocytes were treated with four d… Show more

“…To assess the genetic damage induced by physical and chemical agents including pesticides, various test systems have been described in bacteria, in mammalian cells in vivo and in vitro and in plants [1214]. Arguably, the most reliable genotoxicity evaluation for human health risk is conducted in mammals by the induction of chromosomal aberrations (CAs) and micronuclei (MN).…”

“…A decrease in mitotic index has also been reported to many other pesticides such as etoxazole, karathane LC and sodium arsenite [26][27][28]. However, this fungicide did not affect the RI value.…”

“…In addition, in human lymphocytes, the lowest dose induced significant increase in both abnormal cells (%) and CA/cells. These are chromatid break, fragment, sister chromatid union, dicentric chromosomes, gap, ring chromosomes, chromatid exchanges and polyploidy [27]. The frequency of structural chromosome aberrations and CA/cell ratio were higher in human lymphocyte than those of mouse bone marrow cells, after the treatment of Conan 5FL.…”

“…Pesticides are very reactive compounds forming covalent bonds with various cellular biomolecules and, consequently, can damage cell structures and/or interfere with metabolic processes. Several studies have been conducted on different animal models and cellular systems to investigate the genotoxic effects of pesticides (Bolognesi, 2003;Çelik et al, 2004;Poletta et al, 2009). The induction of CAs formation by exposition to different pesticides has been reported in several different test systems (Bolognesi, 2003).…”
