In Vitro Elution Characteristics of Gentamicin and Vancomycin from Synthetic Bone Graft Substitutes

Abstract: Objects:Beta tricalciumphosphate pellets loaded with individualized antibiotics may represent novel options in the treatment of osteomyelitis and infectious bone disease. Here, the in vitro antibiotic elution of vancomycin and gentamicin from the synthetic bone graft substitutes Cerasorb® and Cerasorb M® was tested. Methods:Antibiotic elution and concentration of gentamcin and vancomycin were measured using photometrically-based measurement and homogeneous particle-enhanced turbidimetric inhibition immunoassay… Show more

“…The reported antibiotic elution profiles of different bioceramics remain fairly similar, with the majority of carriers delivering antibiotics above the minimum inhibitory concentrations for between 3-4 weeks. The two principle exceptions to this are the studies of Rauschmann et al 57 and Maier et al 58 , where the antibiotic elution was significantly shorter, tailing off at 10 and 6 days respectively. This difference might be explained by the fact that in these papers, the antibiotics were added to the pre-formed ceramic pellets rather that during the ceramic mixing phase before final setting.…”

“…Maier et al 89 compared the beta tricalcium phosphates Cerasorb ® and Cerasorb ® M as antibiotic carrier systems impregnated with gentamycin and vancomycin. They reported elution rates for both materials over the MIC of S. aureus.…”
