In vitro strategies for Passiflora have been developed owing to its economic and ecological importance. However, plantlet regeneration through somatic embryogenesis has presented some problems, such as the reproducibility of the protocol and formation of abnormal embryos and plantlets. Thus, this study aimed to establish a protocol exploring the embryogenic potential of immature zygotic embryos (IZE) of the wild species Passiflora miniata Vanderpl. and Passiflora speciosa Gardn. Friable calli, which formed on the abaxial surface of the cotyledons, yielded globular, heart-shaped, torpedo and cotyledonary somatic embryos, characterising the embryogenic response as asynchronous. A high percentage of normal regenerants (90 %) was obtained from IZE in media lacking 2,4-dichlorophenoxyacetic acid (2,4-D) in comparison to the value of normal plantlets (60 %) regenerated from mature zygotic embryos inoculated in media with 2,4-D. This result demonstrates that IZE of P. miniata and P. speciosa possess sufficient levels of endogenous phytohormones to trigger a high rate of indirect somatic embryogenesis. All regenerated plantlets had the same genome size and chromosome number as the explant donor plants. Therefore, the indirect embryogenic pathway, employing IZE inoculated into media free of growth regulators, did not cause changes in the karyotype and morphology. Based on these results, IZE should be considered as explant for the establishment of somatic embryogenesis in other species. Besides, a new, reliable and relatively rapid protocol to recover plantlets of P. miniata and P. speciosa yielded several plants, which were acclimatised and used for ornamental purposes and breeding programs, and for reintroduction into biological reserves.

Cross-talk between phytohormones and sugars is intensely involved in plant metabolism, growth and regeneration. We documented alterations in cytokinin (CK) homeostasis in four developmental stages during de novo shoot organogenesis (DNSO) of kohlrabi (Brassica oleracea var. gongylodes cv. Vienna Purple) seedlings induced by exogenous CKs, trans-zeatin (transZ) and thidiazuron (TDZ), added together with elevated sucrose concentration (6% and 9%). Significant impact of CK and sucrose treatment and their interaction was recorded in all investigated stages, including plantlet development before calli formation (T1 and T2), calli formation (T3) and shoot regeneration (T4). Results showed remarkable increase in total CK levels for transZ treatment, particularly with 9% sucrose. This trend was observed for all physiological and structural groups of CKs. Application of TDZ contributed to little or no increase in CK levels regardless of sucrose concentration. Analysis of expression profiles of organogenesis-related genes involved in auxin transport, CK response, shoot apical meristem formation and cell division revealed that higher sugar concentration significantly downregulated the analysed genes, particularly in T3. This continued on TDZ, but transZ induced an opposite effect with 9% sucrose in T4, increasing gene activity. Our results demonstrated that phytohormone metabolism might be triggered by sucrose signalling in kohlrabi DNSO.

Brassica rapa (AA, 2n = 20) is a prime oilseed species in Bangladesh that can be improved via biotechnological approaches. Rescue of developing embryos after an interspecific hybridization, which otherwise usually tend to abort, is very important for improvement of this species, that require an efficient embryo culture protocol. The immature torpedo shaped embryos (451-700 µm) and mature walking-stick type (>700 µm) embryos of five B. rapa varieties namely, Safal, Agrani, BINA Sarisha-6, BARI Sarisha-6, BARI Sarisha-15 were cultured in basal MS media supplemented with 1 mgL-1 BAP, 0.5 mgL-1 NAA & 1 mgL-1 2,4-D. The immature embryo culture followed indirect somatic embryogenesis process but the mature embryos followed direct organogenesis. The immature embryos induced callus within 11-15 days whereas the mature embryos regenerated plantlets through direct organogenesis within 8-11 days. The varieties Agrani, BARI Sarisha-6 and BINA Sarisha-6 induced the highest percentages of callus from immature embryos and the varieties Safal and BARI Sarisha-15 regenerated the highest percentage of plantlet from mature embryos. Safal and Agrani induced callus and regenerated plantlets comparatively earlier than other varieties. An increase in the concentration of BAP from 10 to 20 ppm in MS media + 5 ppm IBA decreased the size of the plantlets of all varieties except the Agrani. The data could be used in improving the existing embryo rescue protocols. The direct organogenesis process of mature embryos can potentially shorten the breeding cycles.
