In vitro plant regeneration of 4 Capsicum spp. genotypes using different explant types

Abstract: This study evaluates the effectiveness of in vitro regeneration of 4 genotypes of pepper (Capsicum spp.) that differ in origin and functional properties: California Wonder, (ATZ × Sono)F 1 , Jalapeno, and SF-9. In order to investigate the effect of the initial explant type, photoperiod, and presence of 2.0 mg L -1 of glycine in the medium, organogenesis was induced on the MS medium with 2.0 g L -1 2-(N-morpholine) ethanesulfonic acid, 1.7 mg L -1 AgNO 3 , 0.4 mg L -1 indole-3-acetic acid (IAA), and 6.0 mg L -1… Show more

“…Hormones are mostly used after seedling is developed also in MS basal culture medium, not intending to reduce growth time, but to induce multiple shoot bud, callus formation, plant regeneration, flower induction, fruit development and rooting. Considerations for seed viability in germination rate or even seeds pretreatment, which can trigger or enhance germination, were not still reported (Bora et al, 2014;Manzur et al, 2013;Orlinska & Nowaczyk, 2015;Verma et al, 2013).…”

“…These studies have resulted in different protocols and observed a significant diversity of morphogenetic responses dependent on genotype, the explant type and the culture medium conditions (Akram, Gaafar, Safwat, & Diab, 2011;Bora et al, 2014;Gogoi, Acharjee, & Devi, 2014;Grozeva, Rodeva, & Kintzios, 2009;Manzur et al, 2013;Orlinska & Nowaczyk, 2015;Raj, Glint, & Babu, 2015;Valadez-Bustos et al, 2009;Verma, Dhiman, & Srivastava, 2013). Among the various types of explants for the initial establishment of in vitro cultures, the seeds have advantages such as rapid growth, rapid availability of material for subsequent cultivation stages.…”

“…Among the overexpression transformants, many showed an altered fruit colour phenotype, varying from the red of the parental Balady and to bright orange of Topepo rosso. The low efficiency of plant regeneration and transformation by pepper cultures has been widely reported (Steinitz et al, 1999;Li et al, 2003;Sharma et al, 2006;El Nagar 2012;Verma et al, 2013;Orlinska and Nowaczyk, 2015).…”

“…It is the world's most important vegetable after tomato and used as fresh, dried or processed products, as vegetables and spices or condiments (Acquaah, 2004). Its nutritional properties including antioxidants are important for human nutrition (Mateos et al, 2003;Orlinska and Nowaczyk, 2015), phenolics "flavonoids" (Bae et al, 2012), carotenoids (Ha et al, 2007), vitamin C, vitamin E (Garcia-Closas et al, 2004) and alkaloids (Srinivas et al, 2009), which play an important roles in human health. Moreover, pepper is also a source for a natural colors and as medicine (Valadez-Bustos et al, 2009 andZhuang, et al, 2012).…”
