In Vitro Propagation of Heliconia psittacorum by Bud Culture

Abstract: A protocol was developed for in vitro propagation of Heliconia psittacorum L.f. by culture of terminal and axillary buds of rhizomes. Cultures were initiated on modified Murashige and Skoog (MS) medium containing 40 μm BA, 150 ml coconut water/liter, 30 g sucrose/liter, and 2 g Gelrite/liter. Shoot multiplication was achieved on the above medium without coconut water, but supplemented with 10 μm BA. Shoots were rooted on MS basal medium a… Show more

“…Out of 319 calluses cultured for regeneration, 20 transformants were confirmed by PCR and genomic Southern analysis, indicating that the transformation efficiency was 6.6% using the protocol established here. Since MS basal medium is commonly used for root formation and plantlet regeneration in several species (Nathan et al 1992;Rao et al 1996), we A Kanamycin-resistant calluses produced from hypocotyl segments (8 weeks after cocultivation). B Development of kanamycin-resistant shoots (6 weeks after transferring the callus in A to MS basal medium).…”
