In vitro propagation of limonium wrightii (Hance) Ktze. (Plumbaginaceae), an ethnomedicinal plant, from shoot-tip, leaf- and inflorescence-node explants

Abstract: Rapid in vitro propagation of Limonium wrightii (Hance) Ktze. (Plumbaginaceae), an endangered medicinal plant, was achieved by culturing the shoot-tip (primary and lateral), leaf-and inflorescence-node explants. MS (Murashige and Skoog, 1962) medium supplemented with 8.87 mM N 6 -benzyladenine (BA) and 1.07 mM a-naphthaleneacetic acid (NAA) supported induction of adventitious shoots from the shoot-tip, inflorescence-node and middle and basal parts of leaf explants after 60 d of culture. Adventitious shoots wer… Show more

“…Half-strength hormone-free MS medium failed to induce roots from excised shoots after 30 days of culture. Stimulation of rooting by IBA in the present study agrees with similar findings in other medicinal plant species, including Ilex aquifolium (Majada et al 2000), Limonium wrightii (Huang et al 2000), Salvia brachyodon (Misic et al 2006), Nolina recurvata (Bettaieb et al 2008) and Vigna unguiculata (Aasim et al 2009). Plantlets with four to six fully expanded leaves and welldeveloped roots were successfully acclimatized and eventually established in soil (Fig.…”

“…Multiple shoot regeneration from E. alba nodal explants cultured on MS media supplemented with various concentrations and combinations of BAP, NAA and IAA were studied. The effect of BAP in combination with an auxin has been reported and in most of the cases BAP and NAA were used for the induction of multiple shoots of various medicinal plants (Sudha et al, 1998, Huang et al, 2000, Chen et al, 2001, Biswas et at., 2009). The percentage of responded explants, number of shoots per explants, number of leaves, length of shoots and proliferation of shoots differed with the medium constituents.…”

“…Tissue Culture Plants which were used as a source of explants for in vitro studies were grown under controlled conditions and explants were taken from actively growing shoots as described in earlier studies with Limonium wrightii 24) and Adenophora triphylla. 25) To induce multiple shoots, stem-internode, stem-node, shoot-tip and leaf-base explants of S. yoshimurae were cultured on MS basal medium with 4.44 mM BA and 1.07 mM NAA (Table 1).…”

“…A medium supplemented with BA and NAA has been used earlier for in vitro induction of shoots in some medicinal plants. 15,24,25) In S. yoshimurae, multiple shoots were induced not only from pre-existing meristems of the shoot-tip and stem-node explants but also developed from the cut end of the stem-internode and leaf-base explants without an intervening callus phase (Table 1, Fig. 2).…”
