In vitro regeneration of wild chervil (Anthriscus sylvestris L.)

Abstract: Anthriscus sylvestris (L.) Hoffm. (Apiaceae) is a common wild plant that accumulates the lignan deoxypodophyllotoxin. Deoxypodophyllotoxin can be hydroxylated at the C-7 position in recombinant organisms yielding podophyllotoxin, which is used as a semi-synthetic precursor for the anticancer drugs, etoposide phosphate and teniposide. As in vitro regeneration of A. sylvestris has not yet been reported, development of a regeneration protocol for A. sylvestris would be useful as a micropropagation tool and for me… Show more

“…The lowest tested concentration used (0.1 mg l -1 zeatin) provided the highest number of shoots per plant from all 26 tested media. Superior effect of zeatin over kinetin and BA had been reported also for micropropagation of other species (Peixe at al., 2007;Hendrawati et al, 2012). Although zeatin is very expensive chemical compound, it is the only growth regulator capable of inducing satisfactory growth and multiplication, and thus it is not possible to replace it by another cytokinin.…”

“…Plant tissue culture, or the aseptic culture of cells, tissues, organs, and their components under defined physical and chemical conditions in vitro , is an important tool in both basic and applied studies as well as in commercial application [117]. Calli of A. sylvestris did not produce deoxypodophyllotoxin and whole plants were required in order to achieve the differentiated forms to produce deoxypodophyllotoxin for metabolic engineering purposes [118]. In the cell suspension cultures only trace amounts of deoxypodophyllotoxin were detected.…”

“…The substrates usually used for acclimatization with other members of that family (Sharma and Wakhlu 2001, Nath and Buragohain 2003, Tavares et al 2010a, Coste et al 2012, Hendrawati et al 2012, Jaheduzzaman et al 2012 were far different from the substrate which promoted acclimatization of our plant species. H. pastinacifolia did not acclimatize at all if the substrates were rich in organic matter (data not shown).…”
