In vitro screening and isolation of human aromatase inhibitors from Cicer arietinum by a novel continuous online method combining chromatographic techniques

Abstract: Ultrafiltration liquid chromatography with mass spectrometry can efficiently and rapidly screen and identify ligands from the seeds of Cicer arietinum for human aromatase. Using this method, we identified 11 major compounds, including organic acids, organic acid glycosides, flavone glycosides, isoflavones, and isoflavone glycosides, as potent human aromatase inhibitors. A continuous online method, including pressurized liquid extraction, countercurrent chromatography, and preparative liquid chromatography, was… Show more

“…Furthermore, the extract obtained from the second segment had the highest caffeine and theobromine, two compounds of significant bioactivity. Zhang et al [ 190 ] developed HPLE merged with countercurrent chromatography (CCC) and preparative liquid chromatography (PLC) to extract and isolate Cicer arietinum seeds’ phenolics. The HPLE extraction was conducted using an aqueous ethanol solution with a volume-to- volume ratio of 60%.…”

“…PLE was also used in online extraction and separation systems for the extraction of polyphenolic compounds. In this sense, Zhang et al [62] assembled a continuous online system comprising PLE coupled to countercurrent chromatography (CCC) and preparative liquid chromatography (PLC) for the extraction and isolation of bioactive compounds (human aromatase inhibitors) from Cicer arietinum seeds. The PLE extraction was performed using aqueous ethanol (60% v/v) at 80°C for 5 min.…”

“…Hence, semi‐PLC and CCC can be combined to purify the target bioactive compounds with different polarities because of their complementary and orthogonal properties . The combined use of semi‐PLC and other separation techniques is also reported . For the automation of mixed chromatography and integrated separation and extraction of natural compounds, online hyphenated techniques including PLE, semi‐PLC, and CCC were established in this study for the extraction and online isolation of COX‐2 inhibitors from P. amurense Ruprecht.…”
