IN VITRO SHOOT REGENERATION OF INDONESIAN BANANAS (Musa spp.) cv. AMBON KUNING AND RAJA BULU, PLANTLET ACCLIMATIZATIONAND FIELD PERFORMANCE

Abstract: The aim of this study was to observe responses of two banana cultivars 'Ambon' and 'Raja Bulu' on different BA concentrations and effects of different media and fertilizer on survival and growth of plantlets. Sterilized explants were cultured on initiation medium (MS with 1mg L -1 BA) for 4 weeks, then subjected to media MS with 2.5, 5.0, or 7.5 mg L -1 BA. Numbers of shoot buds, shoots and propagules were recorded after 4 consecutive passages with 4 weeks intervals. Rooted plantlets were acclimatized in three… Show more

“…Each cultivar appears to be reactive to an optimal concentration of benzylaminopurine, beyond which the rate of proliferation cannot be increased and may even decrease. Concerning the stem meristematic explants, the concentration of 22.2 μM was indicated by several studies as optimal for the proliferation of most banana and plantain cultivars [42], [41], [45], [46]. Though, other values have been reported in other studies (Table I).…”

“…However, many diseases, such as black sigatoka, fusarium wilt, banana bunchy top virus, burrowing nematodes, and banana weevil borer [15], are associated with traditional propagation techniques [16][17][18]. Therefore, micropropagation has become a common practice for the production of bananas, thus assisting with the production of clean, disease-free plant material [16,19,20]. The tissue culture of bananas also allows large-scale mass in vitro clonal propagation of elite banana varieties, free of diseases.…”
