In Vitro Study of Influence of Au Nanoparticles on HT29 and SPEV Cell Lines

Abstract: Cell culture models are excellent tools for potential toxicity of nanoparticles and fundamental investigations in cancer research. Thus, information about AuNP potential toxicity and effects on human health is necessary for the use of nanomaterials in clinical settings. The aim of our research is to examine the effects of AuNPs on the epithelial origin cell lines: continuous and oncogenic. Embryonic porcine kidney epithelial inoculated (SPEV) cell line and colorectal carcinoma cell line (HT29) were used. In th… Show more

“…Subpopulation of repopulating cells has been identified as the main cause of radiotherapy failure. GNP ability to slow down the cell proliferation goes hand in hand with the cell cycle arrest and has been demonstrated by different groups [26,32,76,108,111]. We reported a 1.4-fold increase in doubling time when A549 cells are incubated with 10 nm GNPs [26].…”

“…Polyvinylpyrrolidone (PVP)-coated GNPs (7.4 nm ± 2.8 nm) activated caspase 3 and enhanced the ratio of apoptotic cells, as seen by annexin V/PI staining in HT-29 cells [ 59 ]. Citrate-coated GNPs (8, 15, and 40 nm) were observed to elicit apoptosis in HT29, SPEV, HL7702, and A549 cells [ 60 , 61 , 62 ]. In a comparative study, citrate-coated GNPs (40 nm) enhanced caspase 3 and Bax gene expression in a dose-dependent manner (4–1600 μg/L) in the head kidney of Sparus aurata , while PVP-coated GNPs (40 nm) showed a U-shaped response, with the highest level of caspase 3 and Bax gene expression induced at 80 μg/L [ 63 ].…”

“…This work addresses, for the first time, a comprehensive study about the traffic of iron and other elements in the subcellular fractions, the integrity of the mtDNA, the mitochondrial functionality and the induction of the autophagy process that will increase the knowledge about the metabolism of nanoparticles and its possible application in therapy focused on mitochondria. To perform the study HT-29 cell line was chosen based on previous studies in which the implications of nanomaterials in therapy have been evaluated [ 13 , 14 , 15 ].…”
