In Vitro Synthesis of Peptidoglycan by β-Lactam-Sensitive and -Resistant Strains of Neisseria gonorrhoeae : Effects of β-Lactam and Other Antibiotics

Abstract: The synthesis in vitro of peptidoglycan by Neisseria gonorrhoeae was studied in organisms made permeable to nucleotide precursors by treatment with ether.Optimum synthesis occurred at 300C in tris(hydroxymethyl)aminomethane-maleate buffer (0.05 M; pH 6) in the presence of 20 mM Mg2e. The incorporation from uridine 5'-diphosphate-N-acetyl-['4C]glucosamine into peptidoglycan, measured after precipitation of the cells with trichloroacetic acid, was sensitive to the fl-lactam antibiotics, bacitracin, diumycin, and… Show more

“…Neisseria gonorrhoeae strain 1 L260 (Brown & Perkins, 1979), grown overnight on chocolate agar plates, was used to inoculate proteose peptone no. 3 (Difco) liquid medium, with pyruvate as carbon source (Blundell & Perkins, 1981).…”

“…N . gonorrhoeae strain IL260, described previously (Brown & Perkins, 1979), grown on chocolate agar plates overnight, was used to inoculate pyruvate-supplemented proteose peptone no. 3 (Blundell & Perkins, 1981) to an OD67S of 0.04.…”

“…Nei.veria gonorrhoeae strain 1 L260 is a penicillin-sensitive isolate (Brown & Perkins, 1979) and the highly autolytic strain RD5 was obtained from R. S. Rosenthal (Rosenthal, 1979). Peptidoglycan was labelled in the glycan backbone by using D-[ 1 -lJC]glucosamine as previously described, and in one experiment penicillin G was present during growth (Blundell & Perkins,198 I).…”
