Depression is a pervasive psychiatric disorder worldwide and diarrhea is considered a leading cause of preventable death for infants and children in developing countries. The current study was done to assess in vivo activities of crude methanol extract of Stixis suaveolens fruit, focusing on antidepressant and antidiarrheal effects. The effect on central nervous system (CNS) was examined by recording the onset of sleep and total sleeping time. Besides, castor oil-induced diarrhea in the mice model was followed while evaluating the antidiarrheal effects. Significant (p<0.01) reduction in onset time (64.8 min) as well as in total sleeping time (64.8 min) were recorded between the test groups and the control. The extract, when given orally, also resulted in a significant (p < 0.001) reduction of diarrheal feces induced by castor oil by 71.43%. The in vivo bioassays demonstrated that the crude methanol extract of the fruit of S. suaveolens contains significant antidiarrheal and CNS stimulating actions. However, further vigorous studies are recommended to isolate and characterize the major phytoconstituents from the plant in order to develop the exact mechanism of actions of the plant against various disorders. Leaves of Bruguiera gymnorhiza (L.) Lam. are very popular traditional remedies used by tribal people of Bangladesh near to Sundarbans. The purpose of this work was to assess the pharmacological attributes of methanolic extract of leaves of B. gymnorrhiza employing mice model. Using the well-known tail tipping method, the in vivo glucose-lowering ability of methanolic extract of leaves of B. gymnorrhiza was investigated. The writhing experiment was performed to determine peripheral analgesic activity. The anti-diarrheal activity was determined utilizing the castor oil-induced diarrhea in the mice model. In consequence, oral treatment of the extract at 400 mg/kg b.w. dose caused significant suppression of acetic acid-induced writhing (58.33%). The findings were comparable to the corresponding standard diclofenac (50 mg/kg b.w.) that exhibited 86.67% inhibition of abdominal writhing. Besides, after 180 min of oral ingestion (400 mg/kg) of the extract, the maximum reduction in blood glucose (38.46%) was found, compared to standard glibenclamide (73.67%). Furthermore, when compared to conventional loperamide (65.06%), there was a 40.02% reduction in diarrheal feces at 400 mg/kg b.w. dose. The methanolic extract of leaves of B. gymnorrhiza has strong peripheral analgesic and anti-diarrheal efficacy, as well as modest glucose-lowering effect, according to these in vivo bioassays. The leaves of Wendlandia paniculata (Roxb.) DC is a rare folk medicine among tribal people even though there is no scientific literature available regarding its pharmacological potentials. This study aimed to evaluate the effects of crude methanol extract of W. paniculata (Roxb.) DC. Leaves in mice model. The in vivo glucose-lowering capacity of methanolic crude extract was analyzed utilizing the well-known tail tipping method. The central and peripheral analgesic activity was evaluated using the ‘tail flick' and writhing assay respectively. The anti-diarrheal activity was evaluated using the method of castor oil-induced diarrhea in mice. As a result, oral administration (400 mg/ kg) of the extract resulted significant (P<0.001) delay in response to pain and inhibition of acetic acid-induced writhing response. The results were comparable to the respective standards morphine (2 mg/kg) and diclofenac (50 mg/kg). Likewise, maximum reduction (p<0.05) of blood glucose (32.3%) was observed 180 min after oral intake(400 mg/kg) of the extract compared to the 47% of the standard glibenclamide. Moreover, a reduction of diarrheal feces was observed 45.83% (p<0.001) compared to standard loperamide 70.83%. The in vivo bioassays confirmed that the crude methanol extract of leaves of W. paniculata possesses significant analgesic (central, peripheral) and antidiarrheal activity with mild glucose-lowering activity. Isolation of bioactive compounds is needed to justify the molecule responsible for the activity.
