In vivo confocal Raman microscopic determination of depth profiles of the stratum corneum lipid organization influenced by application of various oils

Abstract: Both oil types remain in the superficial layers of the SC (0-20% of the SC thickness). Skin treated with mineral- and plant-derived oils shows significantly higher disordered lateral and lamellar packing order of ICL in these layers of the SC compared to intact skin. Plant-derived oils significantly changed the ICL ordering in the depths of 30% and 70-90% of the SC thickness, which is likely due to the penetration of free fatty acids in the deeper layers of the SC.

“…Noninvasive optical methods were employed to analyze the skin penetration of petrolatum and white mineral oil in human subjects (Stamatas et al 2008;Patzelt et al 2012;Choe et al 2014;Choe et al 2015;Choe et al 2017). Test substances were applied to the inner forearm, mainly at dermal loads of 2 mg/cm 2 .…”

“…Petrolatum was exclusively detected on the skin surface. With confocal Raman microscopy, the penetration process can be monitored and the thickness of the horny layer can be determined without the use of a tracer dye (Stamatas et al 2008;Choe et al 2014;Choe et al 2015;Choe et al 2017). The stratum corneum of untreated forearm skin was measured to be 19.2 mm thick (Choe et al 2014;Choe et al 2015).…”

“…For skin analysis not related to cancer, but to characterize depth profiles of stratum corneum, e.g., penetration studies of drugs or cosmetic products, in numerous cases a model 3510 Skin Composition Analyzer (River Diagnostics, Rotterdam, The Netherlands) was frequently employed. [74][75][76][77][78] An alternative miniaturized, confocal fiber optic probe intended to fit within the instrument channel of a standard medical endoscope, with a diameter of 2.8 mm, was developed by Day et al 36 It was optimized for the study of the carcinogenesis process of esophageal malignancy. The optical layout is related to the probe shown in Fig.…”

“…Furthermore, evaluations of intercellular lipids packing and ordering, and SC thickness are commonly performed using CRM [209][210][211][212][213][214], and include applications in skin ageing [211,212] and topical oils [209]. Eklouh-Molinier [211] combined CRM and cutometric data into a Partial Least-Squares (PLS) model in an attempt to profile spectral markers associated with ageing, and used this to create a "biological age" chart that mapped skin age based on its molecular state rather than its chronological age.…”
