In vivo MR quantification of superparamagnetic iron oxide nanoparticle leakage during low‐frequency‐ultrasound‐induced blood–brain barrier opening in swine

Abstract: Purpose: To verify that low-frequency planar ultrasound can be used to disrupt the BBB in large animals, and the usefulness of MRI to quantitatively monitor the delivery of superparamagnetic iron oxide (SPIO) nanoparticles into the disrupted regions.Materials and Methods: Two groups of swine subjected to craniotomy were sonicated with burst lengths of 30 or 100 ms, and one group of experiment was also performed to confirm the ability of 28-kHz sonication to open the BBB transcranially. SPIO nanoparticles were … Show more

“…As is reported, MRI sequences such as T 2 maps can be used to quantify the iron concentration in tissue, making quantification of therapeutic agent delivery easier [26-28]. Here, we noted the linear correlation between the number of labeled cells detected by flow cytometry and corresponding △R2 value (r 2 =0.907, p<0.01; Fig.…”

“…The current and potential usefulness as contrast agents for magnetic resonance imaging and nuclear magnetic resonance the ultra small super magnetic iron oxide gained importance 55 . In MRI guided ultrasound-enhanced brain drug delivery super paramagnetic iron oxide play an important role as theranostic agent 56 . They offer unique properties for cell tracking by magnetic resonance imaging in cellular immunotherapy, subcellular packaging of magnetic nanoparticles 57 .…”

“…Therapeutic anti-amyloid-β antibodies (150 kDa) which were applied to remove Aβ plagues for immunotherapy in Alzheimer's disease treatment have been successfully delivered to the brain 69, 70. Macromolecules such as magnetic nanoparticles (comprising 60-nm SPIO) have also been successfully delivered into the brains of both small and large animals via BBB opening 60, 71, 72. Polyethylene-glycol-coated gold nanoparticles (10-20 nm) have recently been delivered into the normal rat brain, which could have potential photothermal/photoacoustic imaging applications for combined cancer treatment and diagnosis 63.…”
