In Vivo Stimulation of α- and β-Adrenoceptors in Mice Differentially Alters Small RNA Content of Circulating Extracellular Vesicles

Abstract: When myocardial function is compromised as in heart failure (HF), there is activation of the sympathetic nervous system with elevated circulating catecholamine levels. These catecholamines activate cardiac and extra-cardiac adrenergic receptors (ARs). Interest in secreted extracellular vesicles (EVs) from the heart is growing and in HF, it is not known whether excessive activation of α- or β-adrenergic receptors (ARs) could induce specific changes in EV content. In this study, we have evaluated, by next genera… Show more

“…Thus, an organ lesion, although not necessarily one in which a structural change occurs, must illustrate that the organ has developed the lesion. Structural alterations are accumulated in response to a variety of humoral and cellular factors as well as various receptors for prolonged periods [ 6 ]. For myocardial remodeling, because the myocardium is nonrenewable, hypertrophy, apoptosis, and remodeling of the glial network outside the cardiomyocyte can be used as entry points to explain myocardial remodeling.…”

“…Chronic treatment with the β-AR agonist isoproterenol was recently shown to induce cardiac hypertrophy concomitant with a change in the small RNA content of circulating sEVs in mice, though the tissue source of the small extracellular vesicles (sEVs) was not identified. 16 The highlighted work by Lin et al 12 demonstrates that β 3 -AR activity in brown adipocytes does indeed change the makeup of BAT-derived exosomes at the proteome level, through differential packaging of at least iNOS, but the full extent to which β 3 -AR affects the composition of BAT-derived exosomes remains to be seen. A full-scale proteomic and transcriptomic characterization of β 3 -ARdependent packaging of BAT-derived exosomes would certainly be an interesting and worthwhile endeavor.…”
