Increased expression of clp genes in Lactobacillus delbrueckii UFV H2b20 exposed to acid stress and bile salts

Abstract: The ability to survive in harsh environments is an important criterion to select potential probiotics strains. The objective of this study was to identify and carry out phylogenetic and expression analysis by quantitative real-time PCR of the clpP, clpE, clpL and clpX genes in the probiotic strain Lactobacillus delbrueckii UFV H2b20 exposed to the conditions prevailing in the gastrointestinal tract (GIT). Phylogenetic trees reconstructed by Bayesian inference showed that the L. delbrueckii UFV H2b20 clpP, clpL… Show more

“…Exposure of Lb. delbrueckii to 0.1% bile salts increased the expression of clpP and clpE, which, as we have seen above, are involved in repair/degradation of damaged proteins (466). This is in agreement with the observation that mutations in lr1864 (clpL) in Lb.…”

“…delbrueckii cells to acidic conditions prevailing in the GIT (pH 3.5 for 60 min) was shown to result in increased expression of the clpP, clpE, clpL, and clpX genes. As detailed above, the Clp chaperones actively promote refolding or degradation of damaged proteins (466).…”

“…Previous studies have applied the bile stress method or molecular biological method to reveal the mechanism of enhanced bile resistance (Noriega et al 2004;Whitehead et al 2008;Hamon et al 2011;Alc antara and Z uñiga 2012;Ferreira et al 2013). Tween 80 and prebiotics were added to Lactococcus and Lactobacillus cultures to enhance…”

“…The substrate specificity of ClpP is often determined by the regulatory ClpP subunits, and the ClpP subunits alone can function as chaperones. The Clp proteases are found also in several LAB species, and they are important for the survival of some LAB species, including L. casei (33), L. delbrueckii (34), and L. reuteri (35), in acid stress and bile salts. Furthermore, Clp proteases are associated with thermotolerance of the species Strep- (36,37).…”
