Increased Frequency of Pre-germinal Center B Cells and Plasma Cell Precursors in the Blood of Children with Systemic Lupus Erythematosus

Abstract: We have analyzed the blood B cell subpopulations of children with systemic lupus erythematosus (SLE) and healthy controls. We found that the normal recirculating mature B cell pool is composed of four subsets: conventional naive and memory B cells, a novel B cell subset with pregerminal center phenotype (IgD+CD38+centerin+), and a plasma cell precursor subset (CD20−CD19+/lowCD27+/++ CD38++). In SLE patients, naive and memory B cells (CD20+CD38−) are ∼90% reduced, whereas oligoclonal plasma cell precursors are … Show more

“…In addition, the frequency of CD27 + cells was higher in SLE B cells following long-term stimulation. These observations are in agreement with previous studies on SLE reporting higher frequencies of CD27 + blood B cells and plasma cells (Arce et al 2001;Jacobi et al 2003;Odendahl et al 2000), which could be prone to rapid differentiation into Ig-secreting cells through Fig. 3.…”

“…Prosurvival effects may also be associated with the dysregulated overproduction of stimulatory factors, such as interferon-␣ (40), which is currently believed to contribute to the accelerated B cell differentiation into plasma cells seen in pediatric lupus patients (41). Similarly, tumor necrosis factor ␣ and stem cell-derived growth factor 1 may play somewhat similar roles in supporting the erosive synovitis of RA (42,43).…”
