Increased Levels of miRNA-146a in Serum and Histologic Samples of Patients with Uveal Melanoma

Abstract: Purpose: To analyze MiRs expression in serum of UM patients, respect to healthy donors, and to compare this data with MiRs expressed in formalin-fixed, paraffin-embedded UM samples. Methods: Expression profile of 754 miRNAs was performed in serum of patients with uveal melanoma who underwent primary enucleation. The level of miRNAs increased in serum was individually analyzed on FFPE UM samples and compared to choroidal melanocytes from unaffected eyes. Results: Fourteen patients with uveal melanoma were include… Show more

“…It has been established that aberrant expression of miRNAs leads to progression and metastasis of several cancers. In the past decade, several studies have examined the role of microRNAs in pathogenesis and progression of UM by utilizing plasma [24], serum [25], cell lines [26][27][28][29], and clinical tissue specimens [21][22][23]30]. Uveal melanoma is a very rare cancer, making it difficult to obtain a large number of samples from a single institution.…”

“…At the time of metastasis, miR-20a, miR-125b, miR-146a, miR-155 and miR-223 plasma levels had increased, and miR-181a decreased [117]. In serum samples from UM patients undergoing enucleation, eight differentially expressed miRNAs were identified (miR-146a, miR-523 upregulated; miR-19a, miR-30d, miR-127, miR-451, miR-518f and miR-1274B downregulated) compared with normal controls [118]. Furthermore, miR-146a was validated in formalin-fixed paraffin-embedded (FFPE) UM samples and confirmed to be upregulated [118].…”

“…MiR-155 expression is upregulated in lung cancer, and it is positively correlated with the prognosis. [8][9][10][11][12][13][14][15] Therefore, it is speculated that miRNA is likely to be associated with some human tumors, a variety of miRNA expression abnormalities in malignant tumors, but most of the relationship between miRNA and cancer remains to be further studied.…”
