Increased miR-155 Expression in Peripheral Blood Mononuclear Cells of Primary Immune Thrombocytopenia Patients Was Correlated with Serum Cytokine Profiles

Abstract: We investigated the possiblepathogenic role of a microRNA (miR-155) in primary immune thrombocytopenia (ITP). We used quantitative real-time PCR to determine the relative expression of miR-155 and SOCS1 (suppressor of cytokine signaling) mRNA in peripheral blood mononuclear cells (PBMCs) from 28 ITP patients and 28 healthy controls. Cytokine plasma levels were determined by ELISA. Possible associations between miR-155 levels and serum cytokine concentrations were assessed using Spearman or Pearson correlation … Show more

“…As a result, miR-155 [20], miR-409-3p [22], miR-130a [21], and miR-125-5p [57] have gained attention. MiR-155 may contribute to the pathogenesis of ITP by regulating cytokine profiles via SOCS1 [20], while reduced expression of miR409-3p has been detected in ITP patients and leads to up-regulation of IFNG [22]. MiR130A targets TGFB1 and IL-18 and reduced expression of miR130A has also been identified in ITP [21].…”

“…Differentially expressed miRNAs and/or genes were then mapped to the whole network to infer specific miRNA-mRNA subnetworks [12, 20]. According to the framework, stage-specific miRNA-mRNA networks were built, and key miRNAs in the subnetworks were identified on the basis of significant differential expression of miRNAs and genes between each ITP group and the N group using improved POMA [29, 30].…”

“…Increasing evidence has demonstrated that the expression of aberrant miRNAs is associated with the pathogenesis of ITP (13,14). However, the association between miRNAs and the decrease in platelets in patients with ITP was poorly investigated.…”

“…Micro RNAs have also been investigated and it has been shown that increased miR-155 is associated with decreased SOCS1 in ITP. This is thought to be associated with regulation of cytokines given that they also demonstrated the negative correlation of miR-155 and the cytokines IL-4, IL10 and TGF-β1 levels and the positive correlation with IL-17A levels 67 . Interestingly, aberrant miR-155 expression has also been seen in the autoimmune disorders rheumatoid arthritis, systemic lupus erythematous, multiple sclerosis and type 2 diabetes mellitus [68][69][70][71] .…”
