Increased number and frequency of group 3 innate lymphoid cells in nonlesional psoriatic skin

Abstract: These results show that ILCs are present in human skin and indicate that RORγt(+) CD56(+) ILC3 may be involved in the pathogenesis of psoriasis.

“…Skin-derived cells were isolated and analysed after 12 days by flow cytometry after being surface stained, fixed, permeabilized and intracellularly stained. ILC, defined as CD45 + LIN À cells, were gated as previously reported [15]. No statistically significant differences were found in the frequency of CD4 + T cells ( Fig.…”

“…Recent findings have suggested a role for the IL-17A-producing subsets of ILC, namely the group 3 ILC, in the pathogenesis of psoriasis. Although not directly proven to produce IL-17 in vivo, skin-derived ILC3 are increased in the skin of patients with psoriasis [15][16][17].…”

“…IL‐17A production by CCR6 + ILC3 is required for Klebsiella pneumoniae clearance and contributes to protection against systemic Candida albicans infection 53, 123. ILC3‐derived IL‐17A has further been associated with pathogenesis of obesity‐associated airway inflammation and psoriasis in mice and humans 124, 125, 126. Although relatively less characterized, ILC3 also secrete IL‐17F and a role for this cytokine in skin inflammation has also been suggested 127…”

“…For instance, ILC1s, including NK cells, mainly exist in fetal gut and liver; ILC2s are most prevalent in human peripheral blood, lung, and skin; and ILC3s are mostly present in human skin tissue, thymus, tonsils, BM, and gut (6,(15)(16)(17). Due to limited access to these tissues in humans, the function of these tissue-resident human ILCs in healthy and disease statuses has not been well characterized.…”
