Increased plasma level of renin in experimental secondary hyperaldosteronism

Abstract: Renal and peripheral venous plasma was prepared for assay of renin by the technique of Helmer and the formed angiotensin II was assayed for both pressor and steroidogenic activity. Both assays showed higher concentrations of renin in both renal and peripheral venous plasma from dogs with thoracic caval constriction than from normal dogs. The increase in aldosterone and corticosterone secretion by isolated adrenals in response to incubated plasma with its formed angiotensin II was qualitatively similar to that … Show more

“…Plasma renin activity was frequently elevated and the increase for the group was statistically significant; it seems unlikely that the occasional normal value observed was related to the level of sodium intake since this was constant. Also, our earlier studies (12,19) have been done at this level of sodium intake and there was no evidence that it depresses plasma renin activity. Consequently, the findings in dogs and the observations on women suggest that plasma renin activity varies from a normal level to an elevated one during pregnancy.…”

“…When dog renin was added to dog plasma, 94 to 109% was recovered. Evidence for the specificity of the modified Helmer procedure has been presented previously (18,19). Plasma renin substrate was measured by a method described elsewhere (19).…”

“…Each tube contained 0.1 M of disodium ethylenediaminetetraacetate (EDTA) for each 20 ml of whole blood; EDTA served as an anticoagulant and as an inhibitor of angiotensinase activity. Plasma renin was measured by a modified Helmer method adapted for dog blood (18,19); all values are expressed in nanograms of angiotensin II formed during a 4-hour incubation period. When synthetic angiotensin iSupplied by Upl'ohn and Company.…”

“…Samples of renal venous blood were prepared for bioassay by the method of Helmer (22). The pressor response of the rat to angiotensin formed during incubation of the plasma was measured as an index of renin activity as described by Higgins, Davis, Urguhart, and Olichney (23).…”
