Increased production of immune activation marker neopterin by colony‐stimulating factors in gynecological cancer patients

Abstract: Granulocyte colony-stimulating factor (G-CSF) and granulocyte-macrophage colony-stimulating factor (GM-CSF) have recently been introduced in the treatment of chemotherapy-induced neutropenia. Effects of these CSFs on the cellular immune system were evaluated in 38 neutropenic gynecological cancer patients during chemotherapy. In addition to restoring the leukocyte count, GM-CSF--to a greater extent than G-CSF--also induced neopterin, a sensitive marker of macrophages activated by interferons. This effect was c… Show more

“…In patients with gynecological cancer (mostly EOC), Marth et al described an increase in urinary neopterin after treatment with hematopoietic growth factors (101) . An increase in urinary neopterin was detected both in patients treated with GM-CSF and G-CSF.…”

“…G-CSF failed to induce IL-lra protein production in monocytes stimulated with LPS or cultured on adherent IgG (Jenkins and Arend 1993), though neopterin production by the myelomonocytic cell line THP-1 in response to IFN-7 was increased in the presence of G-CSF (Marth et al 1994). Furthermore, in contrast to the other three CSF discussed below, G-CSF did not induce any urokinase-type plasminogen activator (u-PA) activity (Hamilton et al 1991).…”

“…In 18 patients with acute leukemia where G-CSF was administered in the recovery phase during 27 courses of consolidation chemotherapy, peripheral blood monocytes spontaneously produced high concentrations of IL-6 ex vivo (Liu et al 1993). In 38 neutropenic gynecological cancer patients, G-CSF treatment doubled the concentration of serum neopterin (Marth et al 1994), which is regarded as a marker of macrophage activation.…”

“…In addition, we performed serial determinations of serum neopterin levels as a means to measure monocyte/macrophage activation since release of neopterin is a sign of macrophage activation 24,25 and administration of GM-CSF results in increased production of neopterin. 26,27 …”
