Increased vincristine production fromAgrobacterium tumefaciensC58 induced shooty teratomas ofCatharanthus roseusG. Don

Abstract: Dimeric indole alkaloids are used extensively for cancer therapy. Agrobacterium tumefaciens C58 strain was used for induction of shooty teratoma in Catharanthus roseus using epicotyl and stem node explants. The transformed nature of shooty teratomas was confirmed by nopaline assay. Growth kinetics of shooty teratomas depicted maximum growth during 21-24 days of culture. Dimeric alkaloid vincristine in the transformed cultures was present at a concentration of 0.011 that was tenfold higher compared to untransfo… Show more

“…Tenfold higher accumulation of dimeric alkaloid vincristine compared to the untransformed control cultures was reported in the shooty teratoma of Catharanthus roseus which is developed by the infection of A. tumefaciens strain C58 using epicotyl and stem node explants [458]. Similar enhancement effect in terpenoid indole alkaloid production was observed in rol ABC HR lines of C. roseus which developed following the infection with A. tumefaciens strain A4 harboring rol ABC genes [459].…”

“…A poisonous alkaloid chemical compound, Solasodine in shooty teratomas of Solanum eleagnifolium , was reported by Alvarez et al ( 1994 ). In some other plants, shooty teratomas are produced for secondary metabolites production, namely vincristine in Catharanthus roseus G. (Begum et al 2009 ; Begum 2011 ), naphthoquinone in Drosera capensis var. alba (Krolicka et al 2010 ).…”

“…Antidipsotropic activity and herbal remedy uses of daidzin for addiction to alcohol have also been reported (Rooke et al 2000;Lowe et al 2008). Plant tissue and organ cultures are considered as alternative for the supply of bioactive pharmaceutical secondary metabolites (Begum et al 2009;Murthy et al 2014). The hairy roots induced by Agrobacterium rhizogenes infection are genetically stable and grow rapidly in hormone-free media (Giri et al 2003;Chandra & Chandra 2011, Sharma et al 2013Murthy et al 2014).…”
