Inducible nitric oxide synthase response and associated cytokine gene expression in the spleen of mice infected with Clonorchis sinensis

Abstract: Clonorchis sinensis is a food-borne parasite that induces a permanent increase of nitrosation in the body upon infection. The spleen is an important secondary lymphoid organ for the regulation of immune responses locally and in the whole body. However, the functions and mechanisms of the spleen in nitric oxide (NO) responses after C. sinensis infection remain unknown. In this study, BALB/c mice were infected with 20, 40, and 80 C. sinensis metacercariae to simulate mild, moderate, and severe infections, respec… Show more

“…Among the three major isoforms of NOS, inducible NOS (iNOS) is most associated with the pathophysiology of inflammatory diseases [ 7 ]. In our recent studies, we confirmed iNOS induction and associated cytokine expression in BALB/c mice after C. sinensis infection [ 8 , 9 ].…”

“…Amplification and data acquisition were performed with an Applied Biosystems 7500 Real-Time PCR system running on V2.0.6 software (Life Technologies, Carlsbad, CA, USA) and using the following cycling parameters: predenaturation at 95 °C for 30 s, 40 cycles of amplification at 95 °C for 5 s, and annealing and extension at 60 °C for 30 s. Melt curve analysis was performed from 65 °C to 95 °C to determine the melting temperatures (Tm) (Additional file 1 : Table S1) of the identified specific DNA product populations. The mRNA transcript of iNOS was quantified with RT-PCR assay based on the slope of standard curve described previously [ 8 , 30 ]. Serial diluted cDNA samples were used as standards for the preparation of standard curves and acquisition of slopes.…”

“…NO from iNOS-expressing cells suppresses mouse T cell proliferation. Also, in case of deficient iNOS, NO did not produced and this may attributed to increase the infiltration and expansion of CD4 cells (Aheng et al,2011).The spleen was approved to be important organ for iNOS/NO responses and highly expressed iNOS in the splenocytes may regulate Th response negatively (Qing Shen et al,2015).…”

“…In contrast to other constitutively expressed isoforms, iNOS is only expressed in response to inflammatory cytokines such as TNF-α and IL-1β and transcriptional activators, such NF-kB [48, [66][67][68][69]. In chronic inflammation models and inflammation-related tumorigenesis, iNOS may be persistently stimulated by cytokines and NF-kB activation in the tumor microenvironment.…”
